Many modifications lead to changes in the physicochemical properties of proteins, which can lead to inconsistent, underperforming drug products. This is why it’s essential to use a protein services provider you can trust.

Besides deamidation, oxidation and glycosylation, we have strong experience in looking at several other types of post-translational modifications. They can be roughly classified into two groups:

• Process-induced modifications, which may be introduced during sample preparation, down-stream-processing, or other steps involving the use of chemicals, heat or light. Examples are methylation or acetylation of side-chains, fragmentation, and glycation.
• Biologically-induced modifications, which may be introduced in the cell or supernatant by the complex biological system itself. These are often crucial to the correct biological function of the protein. Examples include phosphorylation, sulfatation and formylglycine.

We observe and investigate all modifications by using our fully developed advanced research methodologies, which usually includes RP-HPLC, intact mass determination or peptide mapping. In addition, we can either screen the samples for possible modifications or we can develop a specific test to identify and possibly quantify them.

Characterizing all protein modifications is a key requirement of ICH Q6B.
